One week. Siem Reap (3 nights) → Phnom Penh (2) → Kampot or Kep (2). The classic first trip, and it works because the two long drives are broken by places worth stopping in.
Two weeks. Add the coast properly — two nights on Koh Rong Sanloem after Kampot — and either Battambang between Siem Reap and Phnom Penh, or Kratie on the way north.
Three weeks or more. This is where Mondulkiri and Ratanakiri become realistic. The north-east is a full week on its own by the time you account for the roads.
